Systemic Viral Spread

Systemic viral spread is the dissemination of a virus from its initial site of infection to multiple tissues or organs, often determining disease severity and clinical outcomes. After local replication, viral particles can enter the bloodstream or lymphatic system, producing viremia or lymphatic transport, and then reach susceptible cells according to tissue tropism, receptor availability, and host immune responses. Studying this process helps explain how infections progress beyond their point of entry and how viruses evade or overcome immune defenses. Models of systemic spread support the development of vaccines, antiviral therapies, diagnostic tools, and strategies for limiting transmission and organ damage.

Viral Concentration Determination Through Plaque Assays: Using Traditional and Novel Overlay Systems

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.

A method for surface-spreading chromosomes from budding yeast is presented. This method is derived from a method previously described by Loidl and Klein. In addition, we demonstrate a procedure for immunostaining of spread chromosomes.

Meiosis is the developmental process by which gametes are formed through a single round of DNA replication and two successive rounds of chromosome segregation. Mammalian meiosis can be examined by utilizing a technique to prepare meiotic chromosome spreads. Here, we demonstrate a method of preparing surface-spread nuclei from mouse spermatocytes.
