Mammosphere Enumeration

Mammosphere enumeration is a quantitative method for assessing the number of three-dimensional spheroids formed by mammary epithelial cells in culture, providing an indirect measure of self-renewal and stem or progenitor cell activity. Cells are typically grown under nonadherent, serum-free conditions that limit attachment and favor clonal sphere formation; after a defined incubation period, investigators identify and count structures above a stated size threshold using microscopy or image analysis. In biology, the assay supports studies of mammary gland development, cancer stem-like populations, differentiation, and responses to drugs or genetic perturbations, while consistent seeding density, culture conditions, and counting criteria are essential for meaningful comparisons.
