Membrane Isolation

Membrane isolation is a laboratory method for separating cellular membranes from other cell components so their structure, composition, and function can be studied. To obtain a membrane fraction, cells or tissues are gently homogenized, then centrifuged at increasing speeds to remove intact cells and large debris; density-gradient centrifugation can further resolve organelles according to their buoyant density. The resulting fractions support biochemical analysis of membrane proteins, lipids, receptors, and transport activities, while purity checks help distinguish plasma membranes from intracellular membranes. In biology, this approach clarifies membrane organization, trafficking, signaling, and disease-related changes, and provides material for microscopy, proteomics, and functional assays.

Isolation and Quantification of Axonal mRNAs Using Porous Membrane Inserts and RTddPCR

This study presents a robust method to isolate axonal mRNAs using porous membrane inserts, enabling total neuron vs. neurite separation and RNA purification. Combined with RTddPCR, the approach allows absolute quantification of low-copy transcripts, facilitating studies of mRNA transport and local translation with high sensitivity, reproducibility, and broad experimental applicability.
