Mitochondrial Dynamics Analysis

Mitochondrial dynamics analysis is the study of how mitochondria continually change their shape, size, connectivity, movement, and cellular distribution, revealing how organelles adapt to physiological demands. It typically combines live-cell imaging with quantitative measurements of fission, fusion, morphology, and trafficking; fission commonly involves DRP1-mediated constriction, whereas fusion depends on mitofusins and OPA1 to merge outer and inner membranes. In biology research, these analyses connect mitochondrial architecture with energy production, quality control, apoptosis, and cellular stress, helping investigators evaluate dysfunction in neurodegeneration, metabolic disease, cancer, and other conditions and assess responses to genetic or pharmacological interventions.

Mitochondria play central roles in the regulation of metabolism and homeostasis. Subtle changes in mitochondrial metabolism that affect organismal physiology could be difficult to detect in whole organism metabolomics studies. Here we describe an isolation method that enhances the detection of subtle metabolic shifts in Drosophila melanogaster.

Here we show isolated human platelets can be used as an accessible ex vivo model to study metabolic adaptations in response to the complex I inhibitor rotenone. This approach employs isotopic tracing and relative quantification by liquid chromatography-mass spectrometry and can be applied to a variety of study designs.
