Modified Bouin's Fixative

Modified Bouin's fixative is a specialized chemical solution used to preserve biological tissues for microscopic examination, particularly when clear cellular and nuclear detail is important. Its components act through complementary mechanisms: aldehydes cross-link proteins to stabilize tissue structure, while acidic constituents promote protein coagulation and enhance staining of nuclei and connective tissue. Modified formulations adjust the traditional Bouin's composition to improve compatibility, reduce handling concerns, or suit particular specimens and staining protocols. In biology, this fixative supports histology, developmental studies, and comparative tissue analysis by maintaining morphology during processing, sectioning, and dye-based visualization.

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens. The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper.
