Mosquito Feeding Assay

A mosquito feeding assay is a laboratory method for measuring mosquito feeding behavior, blood-meal uptake, or attraction to a host, making it important for studying disease vectors and evaluating control strategies. In a typical assay, mosquitoes encounter a live host or a membrane feeder containing blood under controlled conditions, and researchers record feeding rate, engorgement, probing, or host preference. These measurements help assess the effects of repellents, attractants, insecticides, and environmental conditions on feeding. The assay also supports research on pathogen transmission by linking mosquito feeding behavior with exposure risk and vector competence.

Behavioral assays provide powerful tools for understanding neuronal function. Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.

Reverse genetic approaches have proven extremely useful for determining which genes underly resistance to vector pathogens in mosquitoes. This video protocol illustrates a method used by the Dimopoulos lab to inject dsRNA into Anopheles gambiae mosquitoes, which harbor the malaria parasite. The technique manipulating the injection setup and injecting dsRNA into the thorax is illustrated.

The standard membrane feeding assay (SMFA) is regarded as the gold standard for the assessment and identification of potential antimalarial compounds. This artificial feeding system is used to infect mosquitoes to further evaluate the effects of such compounds on the intensity and prevalence of the Plasmodium falciparum parasite.
