Msc Surface Markers

MSC surface markers are cell-surface proteins used to identify and characterize mesenchymal stem or stromal cells, which are studied for their capacity to differentiate into bone, cartilage, and fat lineages. Researchers detect these markers by allowing fluorescently labeled antibodies to bind specific membrane proteins, then analyzing the cells through flow cytometry or immunostaining; commonly assessed positive markers include CD73, CD90, and CD105, while negative markers help exclude hematopoietic or endothelial cells. This marker profile supports MSC isolation, quality control, and comparison across cultures, although it should be interpreted alongside morphology, differentiation assays, and other biological characteristics.

Here, a protocol (MitoCeption) is presented to transfer mitochondria, isolated from human mesenchymal stem cells (MSC), to glioblastoma stem cells (GSC), with the goal of studying their biological effects on GSC metabolism and functions. A similar protocol can be adapted to transfer mitochondria between other cell types.

Liver injuries are accompanied by progenitor cell expansion that represents a heterogeneous cell population. Novel classification of this cellular compartment allows for the distinguishing of multiple subsets. The method described here illustrates the flow cytometry analysis and high purity isolation of various subsets that can be used for further assays.

Labeling Neural Cell Surface Markers with Azidosugar while Co-culturing with Endothelial Cells

This video demonstrates a protocol for co-culturing mouse brain endothelial cells and primary cortical stem cells and labeling the cell surface glycoproteins using Ac4ManNAz (azidosugar per-O-acetylated N-azidoacetylmannosamine). This method allows for selective enrichment and identification of cell surface proteins in primary cells.

Mouse embryonic fibroblast can be reprogrammed into induced pluripotent stem cells at low efficiency by the forced expression of transcription factors Oct-4, Sox-2, Klf-4, c-Myc. The rare intermediates of the reprogramming reaction are FACS isolated via labeling with antibodies against cell surface makers Thy-1.2, Ssea-1, and Epcam.

MSC Seeded Scaffold Preparation: A Technique to Prepare Scaffold Seeded with Mesenchymal Stem Cells for Surgical Implantation into Murine Model

2025

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.
