Multiplex Bead Array

Multiplex bead arrays are laboratory assays that measure multiple biological targets simultaneously in a single sample, improving information yield while conserving sample volume. The method uses distinct populations of microspheres, each labeled with a characteristic fluorescent code and coated with a capture molecule; target analytes bind to the appropriate beads, and a fluorescent reporter generates a signal proportional to the amount captured. Specialized analyzers identify each bead population and quantify its associated signal, enabling parallel analysis of cytokines, proteins, antibodies, or nucleic acids. In biology, these arrays support biomarker profiling, immune monitoring, disease research, and high-throughput experimental studies.

Design and Use of Multiplexed Chemostat Arrays

We developed and validated a small-footprint array of miniature chemostats built from readily available parts for low cost. Physiological and experimental evolution results were similar to larger volume chemostats. The ministat array provides a compact, inexpensive, and accessible platform for traditional chemostat experiments, functional genomics, and chemical screening applications.

Analysis of tear film cytokines helps in studying various ocular diseases. Bead based multiplex assays are simple and sensitive and enable the testing of multiple targets in samples with small volumes. Here we describe a protocol for tear film cytokine profiling a using bead based multiplex assay.

This video demonstrates a bead-based multiplex immunoassay technique to simultaneously detect antibodies for multiple environmental pathogens in human saliva. Fluorescent beads were coupled to antigens from different pathogens, which were captured using antibodies in the human saliva sample. Upon labeling the antibodies using a fluorescent reporter, the beads were analyzed using flow cytometry to assess the presence of different pathogen-specific antibodies in the saliva.

This manuscript introduces a robust method of fabricating concave microwells without the need for complex high-cost facilities. Using magnetic force, steel beads, and a through-hole array, several hundred microwells were formed in a 3 cm x 3 cm polydimethylsiloxane (PDMS) substrate.
