Nadh Correction Method

The NADH correction method is an analytical approach used to account for interference from reduced nicotinamide adenine dinucleotide (NADH) in biological measurements, improving the accuracy of assay results. It relies on NADH’s characteristic light absorption, especially near 340 nm, to estimate its contribution to a signal and distinguish that contribution from the analyte or reaction being measured. By applying a measured background or mathematical correction, researchers can interpret enzyme activity, metabolic reactions, and redox changes more reliably. This method is useful in spectrophotometric assays and studies of cellular metabolism, where fluctuations in NADH reflect changes in electron transfer and energy production.

The objective is to monitor the mitochondrial redox state of isolated hearts within the context of physiologic preload and afterload pressures. A biventricular working rabbit heart model is presented. High spatiotemporal resolution fluorescence imaging of NADH is used to monitor the mitochondrial redox state of epicardial tissue.

Here we describe a method to directly visualize microregional tissue hypoxia in the mouse cortex in vivo. It is based on concurrent two-photon imaging of nicotinamide adenine dinucleotide (NADH) and the cortical microcirculation. This method is useful for high resolution analysis of tissue oxygen supply.
