Nasopharynx

The nasopharynx is the upper part of the pharynx located behind the nasal cavity and above the soft palate, where it connects the nose to the throat and supports respiratory function. Air passes through this chamber as its mucosal lining traps particles in mucus, while cilia move the material toward the throat; the nasopharynx also contains the openings of the auditory tubes and lymphoid tissue such as the pharyngeal tonsil. In biology and medicine, studying the nasopharynx helps explain airway conditioning, middle-ear ventilation, immune defense, and the transmission or detection of respiratory infections.

2014

Colonization of the murine nasopharynx with Streptococcus pneumoniae and the subsequent extraction of adherent or recruited cells is described. This technique involves flushing the nasopharynx and collection of the fluid through the nares and is adaptable for various readouts, including differential cell quantification and analysis of mRNA expression in situ.

Processing of Infected Tissues for Bacterial Enumeration in a Mouse Co-Infection Model

(2022)This video demonstrates the processing of lungs from a mouse co-infected with a pathogenic bacterium and a virus. Mincing, homogenizing, and plating the lung tissue allows quantification of bacterial load, indicating bacterial spread to the lungs and disease severity.

A Novel Rescue Technique for Difficult Intubation and Difficult Ventilation

We describe a technique to maintain oxygenation and ventilation using an endotracheal tube inserted nasally to the level of the naso-pharynx while sealing the mouth and nares for successful positive pressure ventilation.

Modeling and Simulations of Olfactory Drug Delivery with Passive and Active Controls of Nasally Inhaled Pharmaceutical Aerosols

This manuscript reviews the modeling and simulations of different protocols to deliver medications to the olfactory region in image-based nasal airway models. Multiple software modules are used to develop the anatomically accurate nose model, generate computational mesh, simulate nasal airflows, and predict particle deposition at the olfactory region.

Intranasal Delivery of mRNA Polyplexes via Rayleigh Breakup Aerosols: An In Vitro Method for Nasal Deposition and Functional Testing

2026

This protocol presents Rayleigh breakup as a mild process for aerosolizing non-viral mRNA vectors. Regional deposition for aerosolized mRNA vectors is evaluated by the Alberta Idealized Nasal Inlet (AINI) model. Green fluorescence protein (GFP) expression in human lung cells represents maintenance of physicochemical and biological functions of post-aerosolized mRNA vectors.
