Neutrophil Viability Assessment

Neutrophil viability assessment is the measurement of whether isolated neutrophils remain alive and functionally intact, an essential step in studies of innate immunity, inflammation, and cellular responses. Common assays evaluate plasma membrane integrity through dye exclusion or detect apoptosis using changes in membrane asymmetry, mitochondrial activity, or nuclear morphology. Researchers use these measurements to distinguish treatment effects from cell damage caused by sample handling, culture conditions, or activation procedures. Reliable viability data support interpretation of neutrophil experiments, including studies of host defense, inflammatory disorders, antimicrobial responses, and drug safety.

We developed an in vitro model that mimics an important component of the acute inflammatory response during infection of the bladder with uropathogenic Escherichia coli. The transuroepithelial neutrophil migration assay enables quantitative assessment of human neutrophil migration across bladder epithelia, cultured on permeable supports, in response to bacterial infection or chemoattractant substances.
