Normalized Enrichment Score

A normalized enrichment score (NES) is a statistical measure that indicates whether a predefined group of biological features, such as genes or proteins, is overrepresented in an ordered dataset relative to a reference expectation. It is typically calculated by comparing the observed enrichment of a gene set with enrichment scores generated from randomized or permuted data, then normalizing for factors such as gene-set size and background distribution. In biology, NES helps identify pathways, functional categories, or molecular signatures associated with a phenotype or experimental condition, enabling comparisons across gene sets and datasets. Positive and negative values generally indicate enrichment at opposite ends of the ranked data.
