Oil-water Fraction Separation

Oil-water fraction separation is a method for dividing an emulsion or mixture of immiscible oil and water into distinct fractions, an important step when biological samples contain hydrophobic and aqueous components. The process relies on differences in polarity and density: because oil does not dissolve in water, the phases coalesce and settle into separate layers, which can then be collected individually. In biology, this approach supports lipid extraction, sample cleanup, and partitioning of hydrophobic molecules before biochemical analysis, helping researchers isolate components and improve measurement of complex biological mixtures.

The low-energy water-accommodated fraction (LEWAF) of crude oil is a challenging system to analyze, because over time, this complex mixture undergoes chemical transformations. This protocol illustrates methods for the preparation of the LEWAF sample and for performing photo-irradiation and chemical analysis by trapped ion mobility spectrometry–FT-ICR MS.

Water in Oil Emulsions: A New System for Assembling Water-soluble Chlorophyll-binding Proteins with Hydrophobic Pigments

This manuscript describes a simple and high-throughput method for assembling water-soluble proteins with hydrophobic pigments that is based on water-in-oil emulsions. We demonstrate the effectiveness of the method in the assembly of native chlorophylls with four variants of recombinant water-soluble-chlorophyll binding proteins (WSCPs) of Brassica plants expressed in E. coli.

Separation and Fractionation of Cell Wall and Cell Membrane Proteins from Mycobacterium tuberculosis for Downstream Protein Analysis

This protocol separates insoluble cell wall and membrane proteins into simple fractions (1-5 proteins) using preparative isoelectric focusing (IEF) based on isoelectric point, followed by separation by molecular weight. The resulting fractions can be used directly for immunological and proteomic analysis without further purification.
