Olfactory Magnetic Tether

An olfactory magnetic tether is a research tool that uses magnetic attachment to restrain or position an animal while its responses to odors are measured. In a typical assay, a lightweight magnetic element connects the subject to a tether, and an external magnetic field controls its location or movement as odorants are presented under defined conditions. This arrangement separates sensory detection from unrestricted locomotion, allowing researchers to quantify odor-guided orientation, attraction, avoidance, or changes in behavioral state. In biology, olfactory magnetic tethers support controlled studies of sensory processing and behavior, linking odor exposure to measurable actions with improved experimental consistency.

Here we describe how to tether a fly in an olfactory magnetic-tether (OMT) apparatus. We describe how to align the rare-earth magnets and odor ports, and how to set mass flow rates for both the stimulus delivery and vacuum suction to achieve optimal odor tracking.

A Simple Flight Mill for the Study of Tethered Flight in Insects

Flight in insects is influenced by a number of factors and the propensity to disperse is an important variable in understanding insect ecology and biological control strategies. We describe the construction and use of a simple, relatively inexpensive, and flexible flight mill for measuring parameters of tethered flight in insects.

Using Insect Electroantennogram Sensors on Autonomous Robots for Olfactory Searches

We describe a protocol for using insect antennae in the form of electroantennograms (EAGs) on autonomous robots. Our experimental design allows stable recordings within a day and resolves individual odor patches up to 10 Hz. The efficiency of EAG sensors for olfactory searches is demonstrated in driving a robot toward an odor source.

Magnetic tweezers, a powerful single-molecule manipulation technique, can be adapted for the direct measurements of the twist (using a configuration called freely-orbiting magnetic tweezers) and torque (using a configuration termed magnetic torque tweezers) in biological macromolecules. Guidelines for performing such measurements are given, including applications to the study of DNA and associated nucleo-protein filaments.
