Optical Density Adjustment

Optical density adjustment is the process of measuring and changing the concentration of suspended biological material so a sample reaches a defined optical density, providing a consistent basis for comparison. A spectrophotometer measures light attenuation, often from scattering by cells, at a selected wavelength; the sample is then diluted with sterile medium or concentrated until its absorbance matches the target value. In biology, this procedure standardizes microbial inocula and cell suspensions before growth experiments, antimicrobial testing, and other assays. Consistent starting densities improve reproducibility and help researchers interpret changes in growth, viability, or treatment response.
