Oxygen Electrode

An oxygen electrode is an electrochemical sensor that measures oxygen concentration or oxygen consumption in liquids, making it useful for studying respiration and metabolism in biology. In a typical Clark electrode, dissolved oxygen passes through a gas-permeable membrane and is reduced at a polarized cathode, generating an electrical current proportional to the oxygen level. Researchers use oxygen electrodes to monitor cellular respiration, mitochondrial activity, photosynthesis, and enzyme reactions under controlled conditions. Measurements of oxygen consumption or production help characterize metabolic pathways, assess tissue and organelle function, and evaluate how biological systems respond to changes in substrates, inhibitors, temperature, or oxygen availability.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

Anaerobic Protein Purification and Kinetic Analysis via Oxygen Electrode for Studying DesB Dioxygenase Activity and Inhibition

Here we present a protocol for anaerobic protein purification, anaerobic protein concentration, and subsequent kinetic characterization using an oxygen electrode system. The method is illustrated using the enzyme DesB, a dioxygenase enzyme which is more stable and active when purified and stored in an anaerobic environment.
