Paneth Cells

Paneth cells are specialized secretory epithelial cells found primarily at the base of intestinal crypts, where they help protect the gut and maintain tissue homeostasis. In response to microbial signals and other stimuli, they release antimicrobial peptides such as defensins and lysozyme, which limit pathogen growth and shape the intestinal microbiota. Paneth cells also support nearby intestinal stem cells by providing niche signals, including Wnt and epidermal growth factor, that promote epithelial renewal. Studying these cells helps clarify host-microbe interactions, intestinal barrier function, and the mechanisms underlying inflammatory and infectious diseases.

Intestinal epithelial stem cells (ISCs) are intermingled with Paneth cells. These cells are differentiated progeny of the ISC, which support the ISCs and provide antibacterial protection. Here we demonstrate how we used transgenic conditional mouse models to establish that Paneth cells play a crucial role in maintaining the intestinal epithelia.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
