Papillae Density Measurement

Papillae density measurement is the quantitative assessment of how many papillae occur within a defined area of biological tissue, most commonly on the tongue, where papillae contribute to surface structure and sensory function. The method typically involves imaging a standardized tissue region, identifying individual papillae through microscopy or digital image analysis, and calculating their number per unit area while controlling for magnification and sampling location. In biology, these measurements support comparisons of tissue morphology among individuals, species, developmental stages, or experimental conditions. They can also help relate structural variation to sensory biology, tissue organization, and changes associated with disease or injury.

Technique to Collect Fungiform (Taste) Papillae from Human Tongue

Knowledge of molecular mechanisms underlying gustatory transduction has recently enjoyed significant advances, largely due to using animal models. However, the wide diversity in taste sensitivity and specificity among mammals warrants studies in human tissue. We describe a biopsy technique to collect living taste cells from the papillae on human tongue.

Isolation and Culture of Human Fungiform Taste Papillae Cells

We aimed to develop a reproducible protocol for isolating and maintaining long-term cultures of human fungiform taste papillae cells. Cells from human fungiform papillae obtained by biopsy were successfully maintained in culture for more than eight passages (12 months) without loss of viability.
