Pcr Genotyping

PCR genotyping is a molecular biology technique used to identify genetic variants or determine an organism’s genotype from DNA, making it valuable for linking inherited sequences to biological traits. The method uses primers designed around a target locus; repeated cycles of denaturation, primer annealing, and polymerase-mediated extension selectively amplify specific alleles, which can then be distinguished by product size, sequence, or fluorescence. In biology, PCR genotyping supports transgene screening, mutant-line identification, population studies, and analysis of genetic variation. Its speed, sensitivity, and requirement for only small DNA samples make it useful for research, diagnostics, and experimental model characterization.

Designer chromosomes of the Synthetic Yeast Genome project, Sc2.0, can be distinguished from their native counterparts using a PCR-based genotyping assay called PCRTagging, which has a presence/absence endpoint. Here we describe a high-throughput real time PCR detection method for PCRTag genotyping.

Frequency and Distribution of Crossovers in Caenorhabditis elegans Meiosis by SNP Genotyping using Real-time PCR

2025

This protocol provides a method for measuring crossover frequencies and distribution during meiosis in Caenorhabditis elegans. Single-nucleotide polymorphism genotyping with real-time PCR was used to determine the origin of the chromosomes. The 4-point mapping allowed to determine the crossover distributions of the left-arm, center, and right-arm regions of each chromosome. ...
