Permanent Marking

Permanent marking is a biological labeling strategy that irreversibly identifies cells or tissues so their persistence, movement, and descendants can be followed over time. In genetic lineage tracing, a site-specific recombinase activates or removes a DNA-based reporter in selected cells, creating a stable mark that is inherited by their progeny during cell division. Researchers use permanent marking to reconstruct cell lineages, map tissue development, measure stem-cell contributions, and determine how cells respond to injury or disease. Because the label remains after the initiating signal ends, this approach connects a cell’s past state with its later location, behavior, and function.
