Phosphate Uptake

Phosphate uptake is the biological process by which cells and organisms acquire inorganic phosphate, an essential nutrient for energy transfer, nucleic acids, membranes, and metabolism. In plants and microorganisms, phosphate moves across cell membranes through specialized phosphate transporters, often powered by proton gradients and regulated according to cellular demand and phosphate availability. Roots can increase acquisition through transporter expression and associations with mycorrhizal fungi, while microbes contribute to phosphate cycling by releasing or absorbing phosphate in soil and aquatic environments. Studying phosphate uptake helps explain nutrient limitation, plant growth, ecosystem productivity, and strategies for improving fertilizer efficiency.

Phosphate Buffer

The phosphate buffer system is a critical biological mechanism for maintaining pH stability in the body. This system operates primarily through two components: sodium dihydrogen phosphate (NaH2PO4), which acts as a weak acid, and sodium hydrogen phosphate (Na2HPO4), which serves as a weak base. Sodium dihydrogen phosphate does not fully dissociate in neutral or acidic solutions. When a strong base, such as sodium hydroxide (NaOH), is introduced into the solution, sodium dihydrogen phosphate...

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

2025

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

Synthesis and Exfoliation of Discotic Zirconium Phosphates to Obtain Colloidal Liquid Crystals

A two dimensional model material of discotic zirconium phosphate was developed. The inorganic crystal with lamellar structure was synthesized by hydrothermal, reflux, and microwave-assisted methods. On exfoliation with organic molecules, layered crystals can be converted to monolayers, and nematic liquid crystal phase was formed at sufficient concentration of monolayers.

Mitochondrial Calcium Uptake Assay: A Plate Reader-Based Method for Measuring Calcium Uptake in Isolated Mitochondria

Source: Cafini, F., et.al. Methodology for the Study of Horizontal Gene Transfer in Staphylococcus aureus. J. Vis. Exp. (2017)This video demonstrates competence-induced DNA uptake in Staphylococcus aureus through natural transformation. A chloramphenicol-sensitive strain engineered to constitutively express a sigma factor is cultured under nutrient-limiting conditions to enhance competence, then incubated with donor DNA carrying a resistance gene. Successful gene transfer is confirmed by the...
