Photon Counting Analysis

Photon counting analysis is a measurement approach that detects and quantifies individual light particles, or photons, to characterize weak optical signals in biological systems. In practice, a photon detector converts each light arrival into an electrical pulse, while counting electronics organize events by time, wavelength, or location and calculate signal intensity and timing. This sensitivity supports fluorescence microscopy, flow cytometry, single-molecule measurements, and luminescence assays when signals are too faint for conventional intensity measurements. By resolving photon arrival patterns, the method can improve detection of rare biological events, distinguish signal from background, and provide quantitative data for studying molecular interactions and cellular processes.

In this article we will describe the procedure for measuring diffusion coefficients using multi-photon fluorescence recovery after photobleaching. We will begin by aligning the laser along the optical path to the sample and determining the proper experimental parameters, then continue generating and finally fitting fluorescence recovery curves.
