Pine Wood Nematode

The pine wood nematode is a microscopic roundworm that causes pine wilt disease, a serious disorder affecting susceptible pine and other conifer species. It spreads primarily when Monochamus beetles carry nematodes to fresh feeding wounds, where the nematodes enter the tree, reproduce, and move through resin canals. Their activity disrupts water transport and contributes to rapid needle discoloration, wilting, and tree death. Studying this biological interaction supports forest health monitoring, laboratory diagnosis, quarantine programs, and research into insect-mediated disease transmission, helping researchers and land managers limit outbreaks and protect economically and ecologically important conifer forests.

Many plant tissues, including phloem and xylem from loblolly pine (Pinus taeda L.), contain high levels of phenolics and polysaccharides that interfere with RNA purification. This presentation discusses techniques for the harvest of field-grown tissues and isolation of RNA of sufficient quality for microarrays and other genomic analyses.

Processing the Loblolly Pine PtGen2 cDNA Microarray

The cDNA microarray PtGen2 was developed for gene expression studies in loblolly pine, P. taeda, and other conifer species. Here, we show pre- and post-hybridization handling and washing techniques that can be used with this array to yield better consistency, reduced artifacts, and lower backgrounds.

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.
