Pma Ionomycin Stimulation

PMA ionomycin stimulation is a laboratory method for activating cells, particularly immune cells, by directly engaging major intracellular signaling pathways. Phorbol 12-myristate 13-acetate (PMA) mimics diacylglycerol to activate protein kinase C, while ionomycin acts as a calcium ionophore that increases cytosolic calcium levels. Together, these signals can activate T cells and other leukocytes while largely bypassing receptor-proximal events, often inducing cytokine production within a defined incubation period. Researchers use this approach to assess cellular responsiveness, measure cytokines by flow cytometry or other assays, and establish positive controls for immune-function experiments.
