Polyq::yfp Reporter

PolyQ::YFP reporter is a genetically encoded fluorescence tool that links a polyglutamine (polyQ) sequence to yellow fluorescent protein (YFP), allowing researchers to visualize polyQ-containing proteins in living cells. The reporter works because the polyQ region can undergo length-dependent conformational changes and self-association, while the YFP tag provides a visible signal whose distribution can reveal diffuse protein, inclusions, or aggregate formation. In biology, this system supports studies of proteostasis, protein misfolding, and neurodegenerative disease mechanisms by enabling microscopy-based analysis of aggregation, cellular localization, and toxicity. It can also help assess genetic or chemical interventions that alter polyQ behavior.

We describe a protocol for visualization of insulin exocytosis in intact islets using pHluorin, a pH-sensitive green fluorescent protein. Isolated islets are infected with adenovirus encoding pHluorin coupled to the vesicle cargo neuropeptide Y. This allows for the detection of insulin granule fusion events by confocal microscopy.
