Ponceau S

Ponceau S is a water-soluble, anionic azo dye used to stain proteins on membranes, making it a rapid way to assess protein transfer and loading in biological experiments. Under acidic conditions, its sulfonate groups interact electrostatically with positively charged amino acid residues, producing visible red bands on nitrocellulose or PVDF membranes. Researchers commonly apply Ponceau S after electrophoretic transfer in Western blotting to evaluate transfer quality, compare total protein distribution, and identify suitable regions for immunodetection. Because the staining is reversible and can be removed by washing, it supports subsequent antibody-based analysis without permanently modifying the membrane.

We report and demonstrate an optimized nitrocellulose binding assay that can be used to quantify autophosphorylation of purified bacterial histidine kinases. Our method has several advantages over traditional SDS-PAGE based techniques, providing a valuable alternative for characterizing these important proteins.
