Post-embedding Protocol

A post-embedding protocol is a set of laboratory procedures performed after a biological specimen has been enclosed in a supporting medium, preparing the embedded sample for analysis. Depending on the embedding material and experimental goal, the workflow may include trimming the block, cutting thin sections, removing or retaining the medium, applying stains or labels, and mounting the sections for microscopy. In biology, these steps preserve tissue architecture while making cellular structures, molecular targets, or ultrastructural features accessible for examination. Proper control of section thickness, surface preparation, and labeling conditions improves image quality, reproducibility, and the interpretation of histological or electron microscopy results.
