Potato Pest Control

Potato pest control is the biological and agricultural management of insects, pathogens, weeds, and other organisms that reduce potato growth, yield, or tuber quality. Effective control combines pest identification and population monitoring with knowledge of pest life cycles, using cultural practices, biological agents, mechanical removal, and carefully selected pesticides when populations exceed action thresholds. These strategies can limit crop damage, slow pesticide resistance, protect beneficial organisms, and reduce environmental impacts. In biology, potato pest control provides a practical example of population regulation, species interactions, plant defense, and integrated pest management in an important food crop.

Agroinfiltration and PVX agroinfection are routine functional assays for transient ectopic expression of genes in plants. These methods are efficient assays in effectoromics strategies (rapid resistance and avirulence gene discovery) and crucial to modern research in molecular plant pathology. They meet the demand for robust high-throughput functional analysis in plants.

Source: Zhao, J., et al. Potato Virus X-Based microRNA Silencing (VbMS) In Potato. J. Vis. Exp. (2020).This video demonstrates the use of Agrobacterium tumefaciens to deliver a Potato Virus X (PVX)-based construct into in vitro-grown potato plants for virus-based microRNA silencing (VbMS). The method enables systemic suppression of a target microRNA, allowing researchers to study its role in plant development through observable phenotypic changes.

This paper describes nitric oxide (NO) fumigation protocols for postharvest pest control. Fumigation chambers are flushed with nitrogen (N2) to establish ultralow oxygen conditions before NO is injected. At the end, chambers are flushed with N2 to dilute NO before exposing products to ambient air to prevent exposure to NO2.

This protocol describes a high through put colorimetric method that relies on the formation of a complex between iodine and chains of glucose molecules in starch. Iodine forms complexes with both amylose and long chains within amylopectin. After the addition of iodine to a starch sample, the maximum absorption of amylose and amylopectin occurs at 620 and 550 nm, respectively. The amylose/amylopectin ratio can be estimated from the ratio of the 620 and 550 nm absorbance values and comparing them...

We present a detailed protocol for potato virus X (PVX)-based microRNA silencing (VbMS) system to functionally characterize endogenous microRNAs (miRNAs) in potato. Target mimic (TM) molecules of miRNA of interest are integrated into the PVX vector and transiently expressed in potato to silence the target miRNA or miRNA family.
