Precursor Rrna Processing

Precursor rRNA processing is the set of molecular steps that converts newly transcribed precursor ribosomal RNA into mature rRNA, an essential stage of ribosome biogenesis and cellular protein synthesis. In this pathway, enzymes and small nucleolar ribonucleoprotein complexes cleave and trim the precursor transcript, chemically modify selected nucleotides, and coordinate its folding with the assembly of ribosomal proteins. These events produce functional ribosomal subunits and ensure accurate translation of messenger RNA. Studying precursor rRNA processing helps explain how cells regulate ribosome production, maintain growth, and respond to cellular stress, while defects in the pathway can disrupt protein synthesis and contribute to disease.

Notch signaling is a form of cellular communication that relies upon direct contact between cells. To properly induce Notch signaling in vitro, Notch ligands must be presented to cells in an immobilized state. We describe an efficient, robust, and cost effective method for extracting nucleic acid from swabs for characterization of bacterial communities using 16S rRNA gene amplicon sequencing. The method allows for a common processing approach for multiple sample types and accommodates a number of downstream analytic processes.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.
