Primitive Heart Tube

The primitive heart tube is the earliest functional form of the vertebrate heart, arising during embryonic development and establishing the basic pathway for blood circulation. It forms when bilateral fields of cardiogenic mesoderm differentiate into cardiac cells, migrate toward the midline, and fuse into a single tube that begins rhythmic contraction; subsequent looping and regional specialization establish the arrangement of future chambers and vessels. Studying this transient structure helps explain how cardiac patterning, blood flow, and heart morphogenesis are coordinated, while providing a framework for investigating congenital heart defects and evaluating developmental effects in experimental models.

Preparation of embryos for Electron Microscopy of the Drosophila embryonic heart tube

We describe a process for fixation, embedding, sectioning, and imaging of late stage Drosophila embryos for Trasmission Electron Microscopy of the embryonic heart tube. This technique allows for the visualization of the heart tube lumen as well as the basement membrane, which lines the lumen of the heart.

Here, we present human pluripotent stem cell (hPSC) culture protocols, used to differentiate hPSCs into CD34+ hematopoietic progenitors. This method uses stage-specific manipulation of canonical WNT signaling to specify cells exclusively to either the definitive or primitive hematopoietic program.

Implantation of Total Artificial Heart in Congenital Heart Disease

This is a case report of a patient with congenitally corrected transposition of the great arteries (CCTGA) who received a total artificial heart (TAH) as a bridge to heart transplant. The TAH was successfully implanted with modifications to accommodate the patient's congenitally malformed heart.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.
