Probenecid

Probenecid is a medication that alters the kidney’s handling of organic acids, making it important in studies of renal transport, drug disposition, and uric acid biology. It primarily inhibits URAT1-mediated reabsorption of urate in the proximal tubule, increasing urinary uric acid excretion, while also affecting organic anion transporters that influence the clearance of several drugs. Clinically, probenecid is used to prevent recurrent gout by lowering serum urate and has been used to prolong the activity of certain antibiotics, including penicillins, by reducing their renal secretion. Its transporter effects also make it a useful experimental tool for investigating membrane transport and pharmacokinetic interactions.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

Unlike that seen for eukaryotes, there is a paucity of studies that detail membrane depolarization and ion concentration changes in bacteria, primarily as their small size makes conventional methods of measurement difficult. Here, we detail protocols for monitoring such events in the significant Gram-positive pathogen Streptococcus pneumoniae utilizing fluorescence techniques.
