Protein Band Detection

Protein band detection is the identification and visualization of discrete protein signals after electrophoretic separation, allowing researchers to determine whether proteins are present and compare their relative abundance or apparent molecular size. In a typical workflow, proteins migrate through a gel according to size and electrical properties, then stains such as Coomassie or silver reveal overall protein patterns, while antibodies can selectively detect a target in a blot. In biology, these measurements support protein purification, gene-expression studies, biomarker research, and validation of recombinant protein production. Careful band interpretation also helps assess sample complexity, separation quality, and experimental consistency.

Ubiquitination is a key posttranslational modification carried out by a set of three enzymes. Mutations of genes involved in this modification are associated with many different human diseases. Here, we describe protocols to detect protein ubiquitination in cultured cells in vivo and test tubes in vitro.

Biotinylated Cell-Penetrating Peptide Probe to Detect Protein-Protein Interactions

This video demonstrates a technique that uses biotinylated cell-penetrating peptides to detect intracellular protein-protein interactions. The cell-penetrating peptides facilitate the conjugated peptide to enter a cell and specifically bind to its interacting proteins. The biotin, after binding to avidin, allows purification of the interacting proteins’ complex from a cell lysate.

Fluorescence Anisotropy-Based Detection of Protein-Protein Interactions

In this video, we describe the fluorescence anisotropy technique to study the interactions between the fluorophore-tagged Shwachman-diamond syndrome (SBDS) protein and the elongation factor-like 1 GTPase (EFL1). On incubating SBDS proteins with gradually increasing concentrations of EFL1, a steady increase in anisotropy is observed, indicating a successful interaction between the two proteins.
