Pyruvate Oxidation

Pyruvate oxidation is the metabolic process that converts pyruvate, the three-carbon product of glycolysis, into acetyl-CoA for entry into the citric acid cycle. In eukaryotic cells, the pyruvate dehydrogenase complex in the mitochondrial matrix removes one carbon as carbon dioxide, transfers electrons to NAD+ to form NADH, and attaches the remaining two-carbon acetyl group to coenzyme A. This irreversible link between glycolysis and aerobic respiration helps regulate carbon flow and energy production. The resulting acetyl-CoA supports ATP generation through subsequent oxidative metabolism, while NADH delivers high-energy electrons to the electron transport chain.

Pyruvate Oxidation

After glycolysis, the charged pyruvate molecules enter the mitochondria via active transport and undergo three enzymatic reactions. These reactions ensure that pyruvate can enter the next metabolic pathway so that energy stored in the pyruvate molecules can be harnessed by the cells. First, the enzyme pyruvate dehydrogenase removes the carboxyl group from pyruvate and releases it as carbon dioxide. The stripped molecule is then oxidized and releases electrons, which are then picked up by NAD+...

This video describes a colorimetric assay for quantitatively measuring pyruvate in the cellular fractions of Caenorhabditis elegans. Utilizing the hydrogen peroxide produced from the enzymatic reaction of pyruvate oxidase with cellular pyruvate, a chromogenic dye is oxidized to produce a colored product, measured using colorimetry to determine the pyruvate concentration.

Fates of Pyruvate

Pyruvate is the end product of glycolysis, where glucose is oxidized to pyruvate, simultaneously reducing NAD+ to NADH. Two molecules of ATP are also produced by substrate-level phosphorylation. In aerobic organisms, pyruvate is metabolized via the citric acid cycle to produce reduced coenzymes NADH and FADH2. These coenzymes are then oxidized in the electron transport chain to produce ATP and, in the process, regenerate the NAD+ and FAD. As seen in some cell types and organisms, fermentation...

Oxidation Numbers

2020

In redox reactions, the transfer of electrons occurs between reacting species. Electron transfer is described by a hypothetical number called the oxidation number (or oxidation state). It represents the effective charge of an atom or element, which is assigned using a set of rules. Oxidation Number (Oxidation State) In the case of an ionic compound, oxidation numbers are assigned based on the number of electrons transferred between reacting species. For example, in the formation of calcium...

Oxidation–Reduction Reactions Earth’s atmosphere contains about 20% molecular oxygen, O2, a chemically reactive gas that plays an essential role in the metabolism of aerobic organisms and in many environmental processes that shape the world. The term oxidation was originally used to describe chemical reactions involving O2, but its meaning has evolved to refer to a broad and important reaction class known as oxidation–reduction (redox) reactions. Some redox reactions involve the transfer of...
