Radiolabeling Yield

Radiolabeling yield is the proportion of a radioactive tracer successfully incorporated into the intended biological molecule or product, making it a key measure of labeling efficiency and experimental quality. It depends on the labeling reaction, including tracer availability, reaction conditions, chemical compatibility, and the separation of labeled product from unreacted isotope or side products; researchers typically quantify the recovered radioactivity relative to the starting material. In biology, radiolabeling yield guides the preparation of probes for tracking DNA, RNA, proteins, metabolites, or receptor-binding compounds. Optimizing yield improves signal strength, reduces waste, and supports reliable studies of molecular transport, metabolism, binding, and cellular function.

Enhancing Efficiency and Radiolabeling Yields of Carbon-11 Radioligands for Clinical Research Using the Loop Method

2024

This protocol presents a detailed procedure to prepare four carbon-11 radiotracers by way of the “loop method” [11C]methylation. We describe the procedure to synthesize [11C]ER-176, with data for three additional radiotracers: [11C]MRB, [11C]mHED, and [11C]PiB. The loop method provides an efficient synthesis with increased radiochemical yields against traditional reaction vessel [11C]methylation.

ATP Yield

2019

Cellular respiration produces 30 - 32 ATP per glucose molecule. Although most of the ATP results from oxidative phosphorylation and the electron transport chain (ETC), 4 ATP are gained beforehand (2 from glycolysis and 2 from the citric acid cycle). The ETC is embedded in the inner mitochondrial membrane and is comprised of four main protein complexes and an ATP synthase. NADH and FADH2 pass electrons to these complexes, which pump protons into the intermembrane space. This distribution of...

The theoretical yield of a reaction is the amount of product estimated to form based on the stoichiometry of the balanced chemical equation. The theoretical yield assumes the complete conversion of the limiting reactant into the desired product. The amount of product that is obtained by performing the reaction is called the actual yield, and it may be less than or (very rarely) equal to the theoretical yield. Percent Yield In the case of chemical reactions, the actual yield of the product is...
