Rat Liver Tissue

Rat liver tissue is a biological model comprising hepatocytes, vascular cells, connective tissue, and bile-duct structures that together perform essential metabolic, detoxification, and secretory functions. Organized into lobules, the tissue receives nutrient-rich blood through the portal vein and oxygenated blood through the hepatic artery; hepatocytes process substrates, modify drugs and toxins through enzyme-mediated reactions, and release bile into canaliculi. Researchers use rat liver tissue to study liver physiology, toxicology, metabolism, regeneration, and disease mechanisms. Fresh, preserved, or cultured samples can reveal cellular responses to chemicals, nutritional changes, injury, and potential therapeutic treatments.

We describe a method to produce an animal model of liver fibrosis in the rat, and assess the degree of fibrosis by histological examination of the liver. The model can be used to study the development of liver disease as well as to test the efficacy of potential anti-fibrotic agents.

A technique for isolating portal fibroblasts from rat liver is described. Livers are perfused and digested in situ with collagenase, followed by ex vivo digestion of the liver slurry and size selection of cells. This method provides a pure population of portal fibroblasts without the need for passage in culture.
