Rescue Plasmid Expression

Rescue plasmid expression is a molecular biology strategy in which a plasmid supplies a functional gene to cells lacking, silencing, or carrying a defective version of that gene, allowing researchers to test whether its activity restores a biological phenotype. After introduction by transfection or transformation, the plasmid’s promoter drives production of the encoded RNA and protein, which can complement the cellular defect under suitable expression conditions. This approach helps establish gene function, distinguish direct genetic effects from secondary changes, and validate mutations or regulatory pathways. Rescue experiments are widely used in cell biology, genetics, disease modeling, and functional studies of engineered organisms.

Capsulorhexis is an important step in phacoemulsification surgery. A surgeon creates a continous curvilinear tear on the anterior lens capsule by controlling the tearing vector forces. A peripherally extended tear is a serious complication. This video demonstrates a novel technique of rescuing capsular radial tear out using a cystotome.

Plasmid Purification

2023

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.
