Rna Folding Isotherms

RNA folding isotherms are quantitative profiles that show how an RNA molecule’s equilibrium structure or folded fraction changes as a solution variable changes at a fixed temperature. They reflect the balance among base pairing, base stacking, tertiary interactions, and stabilization by ions such as magnesium, with changes in salt, ligand, or denaturant conditions shifting the RNA between conformational states. In biology, these measurements help characterize RNA stability, folding pathways, and structural transitions, including those that regulate riboswitches and other functional RNAs. The resulting thermodynamic data can guide structure prediction and clarify how cellular conditions influence RNA activity.

Molecular Chaperones and Protein Folding

The native conformation of a protein is formed by interactions between the side chains of its constituent amino acids. When the amino acids cannot form these interactions, the protein cannot fold by itself and needs chaperones. Notably, chaperones do not relay any additional information required for the folding of polypeptides; the native conformation of a protein is determined solely by its amino acid sequence. Chaperones catalyze protein folding without being a part of the folded protein. The...

A thermodynamic process that occurs at constant temperature is called an isothermal process. Heat slowly flows into the system or out of the system to maintain thermal equilibrium. Processes involving phase changes like water evaporation into steam or freezing water into ice at a constant temperature are examples of Isothermal Processes. An ideal gas can also undergo isothermal expansion or compression. For example, consider 1 mole of an ideal gas inside an isolated cylinder at initial volume V...

Source: Domesle, K. J., et al. Loop-Mediated Isothermal Amplification for Screening Salmonella in Animal Food and Confirming Salmonella from Culture Isolation. J. Vis. Exp. (2020).This video demonstrates the loop-mediated isothermal amplification (LAMP) technique for detecting a target bacterial gene by amplifying DNA at a constant temperature. The reaction mixture is prepared by adding an isothermal master mix, specific primers for the target gene, and samples containing DNA templates. The...

Proteins are chains of amino acids linked together by peptide bonds. Upon synthesis, a protein folds into a three-dimensional conformation which is critical to its biological function. Interactions between its constituent amino acids guide protein folding, and hence the protein structure is primarily dependent on its amino acid sequence.Protein Structure Is Critical to Its Biological FunctionProteins perform a wide range of biological functions such as catalyzing chemical reactions, providing...
