Rna Substrate Generation

RNA substrate generation is the preparation of defined RNA molecules that serve as reactants in biochemical and molecular biology experiments. Typically, a DNA template containing the desired sequence is transcribed in vitro by an RNA polymerase, such as T7 RNA polymerase, and the resulting transcript is purified and prepared under conditions that preserve its intended structure. Researchers use these substrates to measure RNA cleavage, processing, modification, binding, and enzymatic activity, including the function of ribozymes and RNA-associated proteins. Consistent substrate quality and sequence control improve assay reproducibility and help clarify mechanisms of RNA metabolism, regulation, and molecular recognition.

CRISPR/Cas systems mediate adaptive immunity in Bacteria and Archaea. Many Cas proteins are proposed to act as endoribonucleases acting on crRNA precursors of varying length. Here we illustrate three different approaches to generate pre-crRNA substrates for the biochemical analysis of Cas endonuclease activity.

Micropunching Lithography for Generating Micro- and Submicron-patterns on Polymer Substrates

A micropunching lithography approach is developed to generate micro- and submicron-patterns on top, sidewall and bottom surfaces of polymer substrates. It overcomes the obstacles of patterning conducting polymers and generating sidewall patterns. This method allows rapid fabrication of multiple features and is free of aggressive chemistry.

RNA polymerase II synthesizes a precursor RNA that extends beyond the 3' end of the mature mRNA. The end of the mature RNA is generated cotranscriptionally, at a site dictated by RNA sequences, via the endonuclease activity of the cleavage complex. Here, we detail the method to study cleavage reactions in vitro.
