Rodent Oral Models

Rodent oral models are laboratory systems that use mice or rats to study the structure, physiology, and disease processes of the mouth, providing a controlled bridge between basic biology and human oral health research. Investigators alter defined conditions, such as diet, microbial exposure, inflammation, or tissue injury, then assess changes through behavioral measurements, imaging, histology, and molecular analysis. These models help clarify mechanisms of dental caries, periodontal disease, oral infection, wound healing, and bone remodeling while enabling controlled testing of preventive strategies and treatments. Their reproducibility and experimental flexibility make them valuable for linking cellular and immune responses to whole-organism outcomes.

Microhardness is a mechanical property and an informative parameter for evaluating hard tissue pathophysiology. Here, we demonstrate a standardized protocol (sample preparation, polishing, flat surface, and indentation sites) for microhardness analysis in tooth and alveolar bone in rodent oral disease models, namely, dental fluorosis, and ligature-induced periodontal bone resorption.

Oral Infection Alters the Gut Microbiota in a Rat Model

Begin with a rat model with sutures tied around its upper molar teeth to support bacterial colonization.Repeatedly smear the sutures with a gum disease-causing oral pathogenic bacterium using a gel-like solution. The bacteria colonize the gum around the sutures.Colonization triggers inflammation and damages the tissues. The bacteria release toxins that diffuse through damaged tissues and enter the bloodstream. These circulating toxins reach the gut, cause inflammation, disrupt the epithelial...
