Rotary Shadowing

Rotary shadowing is an electron microscopy preparation technique that creates a three-dimensional surface replica of biological specimens by depositing a thin metal layer while the sample rotates. During high-vacuum evaporation, platinum or another heavy metal strikes the specimen from an oblique angle; continuous rotation coats exposed surfaces and reduces directional shadows, while the resulting metal film provides contrast in the electron microscope. The method is used to visualize isolated proteins, nucleic acids, membranes, and cytoskeletal assemblies, especially when their overall shape and spatial organization are more informative than fine molecular detail. Rotary shadowing remains valuable for linking molecular architecture to biological function.

The technique presented here measures the path of freely swimming microscopic species using single wavelength exposure. C. elegans are used to demonstrate shadow imaging as an inexpensive alternative to costly microscopes. This technique can be adapted to accommodate various orientations, environments and species to measure direction, speed, acceleration and forces.
