Semi Thin Sectioning

Semi-thin sectioning is a biological microscopy technique that produces resin-embedded tissue sections thicker than ultrathin electron-microscopy sections but thin enough for detailed light-microscopic analysis. After fixation, dehydration, and resin embedding, a glass or diamond knife mounted on an ultramicrotome cuts sections typically about 0.5–2 micrometers thick, which are collected on slides and stained to reveal cellular structures. This approach preserves tissue architecture while providing high-resolution views of cell boundaries, organelles, and pathological changes. It is widely used in histology, developmental biology, and pathology, and can guide the selection of adjacent regions for subsequent transmission electron microscopy.

We have developed a Semi-automated Optical Heartbeat Analysis method (SOHA) for analyzing high speed optical recordings from Drosophila, zebrafish and embryonic mouse hearts. We demonstrate the application of our methodology to the analysis of heart function in fruit fly and embryonic mouse hearts.
