Sequence Assembly

Sequence assembly is a bioinformatics process that reconstructs longer DNA or RNA sequences from many shorter fragments, enabling researchers to study genomes and transcriptomes when complete sequences are not directly available. Assembly algorithms identify overlaps among sequencing reads or connect shared k-mers in de Bruijn graphs, then merge compatible fragments into contigs and, when possible, scaffolds using paired-end or long-read information. In biology, sequence assembly supports genome annotation, comparative genomics, pathogen surveillance, and the characterization of organisms without reference genomes. Assembly quality depends on read accuracy, coverage, repetitive regions, and the algorithm used, making validation essential for reliable downstream analysis.
