Sodium Glucose Cotransporters

Sodium glucose cotransporters are membrane proteins that use the sodium electrochemical gradient to move glucose across cell membranes, supporting nutrient absorption and blood glucose regulation. In secondary active transport, sodium flows down its concentration gradient through the transporter and drives glucose uptake against its concentration gradient; the sodium gradient is maintained by the Na+/K+-ATPase. SGLT1 primarily enables glucose and galactose absorption in the small intestine, while SGLT2 reabsorbs most filtered glucose in the kidney. Their roles in intestinal physiology, renal function, and glucose homeostasis make them important subjects in biology and targets for therapies that lower blood glucose by promoting urinary glucose excretion.

We present a protocol to screen anti-hepatitis B virus (HBV) compounds targeting pre- and post-viral entry lifecycle stages, using isothermal titration calorimetry to measure binding affinity (KD) with host sodium taurocholate cotransporting polypeptide. Antiviral efficacy was determined through the suppression of viral lifecycle markers (cccDNA formation, transcription, and viral assembly).

Assessing Glucose Uptake in the Motor Neurons of Drosophila Larvae with Upregulated Glucose Metabolism

2025

The video demonstrates a technique for measuring glucose uptake in Drosophila larval motor neurons using a fluorescence resonance energy transfer (FRET)-based intracellular glucose sensor. After dissecting the larvae to expose the ventral nerve cord, the motor neurons are visualized through the fluorescence emitted by the sensor's donor fluorophore. When stimulated with glucose, FRET between the sensor's donor and acceptor fluorophores triggers the emission of fluorescence from the acceptor,...
