Sonication Centrifugation

Sonication centrifugation is a biological sample-preparation workflow that combines ultrasonic cell disruption with centrifugation to break open cells and separate their components. Sonication uses high-frequency sound waves to generate localized cavitation, producing mechanical forces that disrupt cell membranes and release intracellular contents; centrifugation then applies centrifugal force to partition the resulting mixture according to particle size and density. This approach supports cell lysis, tissue homogenization, organelle fractionation, and preparation of clarified extracts for protein analysis, nucleic acid studies, and biochemical assays. Carefully controlling sonication energy, temperature, and centrifugation conditions helps preserve biological molecules and improve sample consistency.
