Sonication Protein Release

Sonication protein release is a cell-disruption method that uses high-frequency sound energy to liberate proteins from biological cells or tissues for downstream analysis. A probe or ultrasonic bath transfers acoustic energy into a liquid, producing cavitation bubbles whose rapid formation and collapse generate shear forces that break cell membranes; cooling helps limit heat-related protein damage. In biology, the resulting lysate can support protein extraction, purification, quantification, and assays such as electrophoresis or immunodetection. Optimizing sonication intensity, pulse duration, sample volume, and temperature improves disruption while helping preserve protein structure and activity.

This video demonstrates the dye release assay for quantifying the enzymatic activity of antimicrobial proteins. The antimicrobial protein degrades the bacteria's dye-labeled cell wall and releases the dye molecules turning the solution blue. The absorbance of the solution correlates with the enzymatic activity of the antimicrobial protein.

Differential diagnostics in painful arthroplasty is crucial for treatment success. Joint aspiration is routinely performed preoperatively. This video demonstrates mast cell exocytosis induced by an activating reagent that includes a calcium ionophore, initiating the release of secretory granules containing the fluorescent reporter protein NPY-mRFP.
