Spectrin Network

The spectrin network is a membrane-associated cytoskeletal framework that supports cell shape, mechanical resilience, and membrane organization. It forms when flexible spectrin heterotetramers connect short actin filaments and attach to membrane proteins through adaptor complexes, creating a deformable lattice beneath the lipid bilayer. In biology, this network is especially important in red blood cells, where it allows repeated passage through narrow blood vessels, and in neurons, where it helps organize specialized membrane domains. Studying spectrin structure and interactions clarifies how cells withstand stress and how defects can contribute to inherited membrane disorders and neurological disease.

Functional Calcium Imaging in Developing Cortical Networks

Spontaneous activity of developing neuronal networks can be measured using AM-ester forms of calcium-sensitive indicator dyes. Changes in intracellular calcium, indicating neuronal activation, are detected as transient changes in indicator fluorescence with one- or two-photon imaging. This protocol can be adapted for a range of developmentally-dependent neuronal networks in vitro.
