Re-insertion Pathway

The re-insertion pathway is a molecular process that places a DNA sequence, such as a mobile genetic element or engineered fragment, back into a genome after it has been removed or mobilized. It typically involves recognition of the DNA ends, processing or excision, selection of a target site, strand transfer, and repair of the resulting junctions by host enzymes. In biology, this pathway helps explain transposon movement, genome rearrangement, and the persistence of genetic elements, while related mechanisms support transposon mutagenesis and genome engineering. Studying re-insertion reveals how cells maintain genome integrity while permitting genetic variation.

The Giant Fiber System is a simple neuronal circuit of adult Drosophila melanogaster containing the largest neurons in the fly. We describe the protocol for monitoring synaptic transmission through this pathway by recording post synaptic potentials in dorsal longitudinal (DLM) and tergotrochanteral (TTM) muscles following direct stimulation of the Giant Fiber interneurons.
