Spermatogonia

Spermatogonia are diploid germ cells located along the basement membrane of the seminiferous tubules, where they initiate and sustain sperm production in the testes. Through regulated mitotic divisions, some spermatogonia self-renew to maintain the stem-cell population, while others differentiate into primary spermatocytes that enter meiosis and ultimately produce haploid sperm cells. Their development depends on interactions with Sertoli cells and hormonal signals within the testicular environment. Studying spermatogonia is important for understanding male fertility, testicular development, reproductive disorders, and the cellular basis of spermatogenesis, with implications for fertility preservation and reproductive medicine.

This work describes the standardization of a method to obtain purified germ cell populations from testicular tissue of different mammalian species. It is a straightforward protocol that combines mechanical testis dissociation, staining with Hoechst-33342 and propidium iodide, and FACS sorting, with wide applications in comparative studies of male reproductive biology.

Germ Cell Transplantation and Testis Tissue Xenografting in Mice

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.

A simple method to derive and maintain spermatogonial stem and progenitor cell lines from adult mice is presented here. The method utilizes feeder cells originating from the somatic cell compartment of the adult mouse testis. This technique is applicable to common mouse strains, including transgenic, knock-out, and knock-in mice.
