Myoepithelial Cells

Myoepithelial cells are contractile epithelial cells located between glandular epithelial cells and the basement membrane, where they help propel secretions through ducts. Their actin- and myosin-rich cytoskeleton contracts in response to stimulation, compressing structures such as mammary, salivary, and sweat gland acini and supporting efficient fluid movement. In biology and tissue research, these cells are studied through their distinctive morphology, molecular markers, and interactions with neighboring epithelial and stromal cells. Assessing myoepithelial organization can clarify gland development, tissue architecture, and changes associated with diseases such as breast cancer.

Isolation of Myoepithelial Cells from Adult Murine Lacrimal and Submandibular Glands

The lacrimal gland (LG) has two cell types expressing α-smooth muscle actin (αSMA): myoepithelial cells (MECs) and pericytes. MECs are of ectodermal origin, found in many glandular tissues, while pericytes are vascular smooth muscle cells of endodermal origin. This protocol isolates MECs and pericytes from murine LGs.

A method to process human mammary surgical discard material is described. Processed tissue, in the form of organoids, can be stored frozen indefinitely or placed in culture for long-term growth. This method enables experimental examination of normal human epithelial cell biology, and the effects of exogenous perturbations.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.
