Squamous Cells

Squamous cells are thin, flat epithelial cells that form coverings and linings throughout the body, where their shape supports either rapid exchange or surface protection. In simple squamous epithelium, a single layer creates a short diffusion distance for gases, nutrients, and wastes, while stratified squamous epithelium uses multiple layers to resist abrasion; surface cells may also produce keratin for added protection. Biology and medical research examine squamous cells in tissues such as blood vessels, alveoli, skin, and the oral cavity. Their appearance, organization, and changes under stress or disease provide important information for histology, cytology, tissue function, and cancer diagnosis.

Perineural invasion (PNI) is a common feature of head and neck squamous cell carcinoma (HNSCC), conferring lower survival rates. Its mechanisms are poorly understood. Utilizing neurites generated from murine dorsal root ganglia confined to a semisolid matrix, the pathways involved in the PNI of HNSCC cell lines can be investigated.

A comprehensive overview of the techniques involved in generating a mouse model of oral cancer and quantitative monitoring of tumor invasion within the tongue through multi-photon microscopy of labeled cells is presented. This system can serve as a useful platform for the molecular assessment and drug efficacy of anti-invasive compounds.
